Looks like playing games and watching sci-fi flicks didn't do the University of Iowa's Jack Scudder any harm. The NASA-funded researcher has been studying elusive magnetic portals connecting the Earth and Sun, and now he's figured out how to find them. The portals, also known as X-points in Scudder-speak, are born from the mingling of Earth's magnetic field with incoming solar winds. These astral connections create flux transfer events (we've got Doc Brown's attention) -- high-energy particle flows responsible for, among other things, the eerie twinkling of the polar auroras. Off the back of Scudder's data wizardry, NASA's planning the 2014 Magnetospheric Multiscale Mission (MMS), sending four craft into the void to observe the portals. Each spacebot is capable of locating them, and when one is found, inviting the others 'round for a study date. Taking a leaf from Scudder's book, Engadget researchers have tracked down a NASA video detailing the mission, located beyond the fold for your convenience.

Syria army presses rebels; Assad regrets shootdown

BEIRUT (Reuters) - The Syrian army pressed its offensive against rebels on Tuesday, bombarding the suburban Damascus city of Douma, while President Bashar al-Assad said he wished his forces had not shot down a Turkish warplane two weeks ago.

The downing of the Turkish F4 in disputed circumstances aggravated tensions between Damascus and Ankara, which responded with high-profile military moves to ward off Syrian helicopters from Turkey's border zone where rebels and refugees are camped.

A Syrian general and 84 soldiers were the latest to defect and flee to Turkey on Monday. But analysts say army and government defections have so far made barely a dent in Assad's armor.

There were few signs that diplomacy might stem Syria's 16-month-old conflict, in which Syrian opposition leaders say more than 15,000 people have been killed. World powers at the weekend made a show of unity by pledging to back talks on a transitional government. But they failed to narrow differences between the West and Russia over Western demands that Assad must go.

Neither Assad's government nor the groups trying to topple him have shown much interest in peaceful compromise. Instead, they seem to be digging in for a winner-take-all struggle.

Anti-Assad activists said there was no let-up in violence across the country.

"There was heavy shelling all morning, now it has calmed down a bit but the siege of the city continues," said Abu Rami, an activist in Homs, the central city which has borne the brunt of the army onslaught against rebel strongholds.

"We are living with little food and little water," he said.

In Damascus suburbs and villages adjacent to Douma, the army shelled pro-opposition areas to which Douma residents fled at the weekend, according to Omar Hamzeh, spokesman for the revolutionary council of rural Damascus.

He said Erbeen, Hamoreyah and Mazare'a came under fire and at least six people were killed during the morning.

EXISTENCE AT STAKE

General Fahd Jassem al-Freij, Chief of the General Staff of the Army and the Armed Forces, was quoted as saying Syria is engaged in war in which conspirators are seeking to destroy its unity and end its existence.

The official news agency SANA quoted him as saying Syria had dealt positively with all Arab and international initiatives to find a settlement, but its enemies had replied with escalation on the ground, coupled with media frenzy to disrupt normal life.

In Cairo, Arab states and Turkey urged Syria's divided opposition on Monday to come together and form a credible alternative to the Assad government, but rifts swiftly emerged.

The talks are being boycotted by the Free Syrian Army, which is leading the armed struggle against Assad. The two-day meeting was organized by the Arab League to try to rally Syria's opposition, whose chronic infighting has scared off potential foreign backers.

"It is not acceptable to waste this opportunity in any way. The sacrifices of the Syrian people are bigger than us all and more precious than any differences or individual and party interests," Arab League Secretary-General Nabil Elaraby said, addressing the roughly 200 Syrian politicians and activists.

ARM "THE REBELS"?

A meeting in Paris this coming Friday of the loose alliance known as the Friends of Syria is likely see the United States in particular come under greater pressure from Turkey and the Syrian opposition's Arab allies - principally Saudi Arabia and the Gulf state of Qatar - to increase aid to the rebels.

Washington has long worried about the wisdom of backing Syria's opposition, which it sees as ill-organized, disparate and much too close to al Qaeda-linked militants.

"We're concerned about pouring more weapons into an already over-militarized situation," U.S. State Department spokeswoman Victoria Nuland said on Monday. "We've made our decision.

Assad meanwhile told a Turkish newspaper he wished Syrian forces had not shot down a Turkish jet on June 22 and he would not allow the tensions to turn into open combat.

"We learned that it (the plane) belonged to Turkey after shooting it down. I say 100 percent 'if only we had not shot it down'," the Cumhuriyet newspaper quoted Assad as saying in an interview published on Tuesday.

Syrian helicopters bombarded Douma on Monday and Turkey scrambled warplanes near the border far to the north, as the U.N. human rights chief warned that arms supplies to the rebels and the government were deepening the conflict.

Assad also said Syria would not mass military forces along the Turkish border, whatever action Turkish Prime Minister Tayyip Erdogan's government took.

Assad also said Syria would not shy away from apologizing if it emerged that the plane was downed in international airspace.

"The plane was using a corridor which Israeli planes have used three times before. Soldiers shot it down because we did not see it on our radar and because information was not given.

"Of course I might have been happy if this had been an Israeli plane," Assad added.

New York-based Human Rights Watch issued a report detailing what it said was "an archipelago" of Syrian state torture centers and the methods used. It included accounts from victims who were beaten with batons and cables, burned with acid, sexually assaulted, and had their fingernails torn out.

The United Nations has not released an estimate of the overall death toll since May, when it said some 10,000 people had been killed. But the Syrian Expatriate Organisation said the figure was now more than 16,000 while Riad Saif of the opposition Syrian National Council put it at more than 20,000, accounting for corpses buried in mass graves and gardens.

Strongest evidence yet of "God particle": U.S. physicists

BATAVIA, Illinois (Reuters) - Physicists at Fermi National Accelerator Lab said Monday that they have come tantalizingly close to proving the existence of the elusive subatomic Higgs boson - dubbed the "God particle" because it brings mass and order to the universe.

But the debris from trillions of collisions between beams of protons and anti-protons over 10 years at Fermilab's now-shuttered Tevatron accelerator outside Chicago still fell short of the scientific threshold proving the discovery. The same collision debris hinting at the existence of the Higgs could also come from other subatomic particles.

The power to confer mass seemed to endow the Higgs with the power of creation itself, which helped lead to its nickname "the God particle."

On Wednesday, physicists at CERN, the European particle accelerator near Geneva, are scheduled to announce their own findings in the Higgs hunt. CERN houses the world's most powerful particle accelerator, the Large Hadron Collider (LHC).

"This is the best answer that is out there at the moment," said Fermilab physicist Rob Roser. "The Tevatron data strongly point toward the existence of the Higgs boson, but it will take results from the experiments at the Large Hadron Collider in Europe to establish a firm discovery."

Physicists not connected to Fermilab also expressed cautious optimism that the long-sought particle had finally deigned to make an appearance.

"These intriguing hints from the Tevatron appear to support the results from the LHC shown at CERN in December," said Dan Tovey, Professor of Particle Physics at the University of Sheffield. "The results are particularly important because they use a completely different and complementary way of searching for the Higgs boson. This gives us more confidence that what we are seeing is really evidence of new physics rather than just a statistical fluke."

No one was ready to declare victory, however. "We will need to wait until Wednesday and the latest results from the LHC before getting the full picture," said Tovey. And although CERN spokesman James Gillies called Fermilab's "a nice result," he quickly added that "it will be interesting to see how it lines up with CERN's results on Wednesday. Nature is the final arbiter so we'll have to be a little more patient before we know for sure whether we've found the Higgs."

Tom LeCompte, a scientist at Argonne National Lab who works at CERN and knows the results, said he was confident the Higgs would be shown to exist, or not exist, this year. But he would not disclose if the findings scheduled to be unveiled Wednesday would be definitive.

"I know the 2012 is the year. I can't tell you July is the month," he said.

Others were less circumspect. "This is the most exciting week in physics history," said theoretical physicist Joe Lykken of Fermilab.

Gene healing in a lotion? Researchers are close

Most people who buy cosmetic lotions and potions know that while the people working behind the department store makeup counters may wear white lab coats, the stuff they sell is more about packaging than science.

But a Northwestern University team is bucking that image, reporting today that they?ve created a way to regulate genes affecting the skin -- merely by applying moisturizer.

Not only could their technology pave the way for cosmetics that actually work, but it also might also prove to be a valuable weapon in fighting melanoma, the deadliest form of skin cancer, or diseases like psoriasis, and wounds like the intractable sores that often plague diabetics.

?This is a blockbuster in the ways we will treat diseases of the skin,? said?Chad Mirkin, director of the International Institute for Nanotechnology and the George B. Rathmann Professor of Chemistry at Northwestern said. ?We?re talking about ailments, scarring, wound healing, ways of regulating them or retarding them.?

In a research paper published today in the Proceedings of the National Academy of Sciences, Mirkin and his colleagues describe not a drug, exactly, but a way of delivering small sections of nucleic acids (DNA and RNA are nucleic acids) called short interfering RNA, or siRNA, to cells. The cells take up the siRNA, which then alters the way a gene inside each cell can be read by the protein-making system.

The team used gold particles with a diameter of 13 nanometers. (One nanometer is 1-billionth of a meter. A typical strand of human hair is roughly 60,000 nanometers wide.) They coated the particles with siRNA to create what they call ?spherical nucleic acid nanoparticle?conjugates,? or SNAs. Millions of SNAs were then added to a commercially available petroleum-based skin moisturizer and the mixture was applied to mice and to lab-grown human skin. ?

In their key experiment in mice, they used their new system to tamp down the activity of a gene called epidermal growth factor receptor, or EGFR, that?s involved in the growth of melanoma. As its name implies, EGFR receives messages from the epidermal growth factor protein. So toning down EGFR will interrupt the message; growth will be reduced or stop.

After mice were treated with the mixture three times per week for three weeks, the expression of the EGFR gene was reduced by 65 percent.

'Impressive' results
Steve Dowdy, professor of cellular and molecular medicine at the University of California San Diego, and a Howard Hughes Medical Institute investigator specializing in RNA inhibition and ways to deliver siRNAs, called that result ?impressive.??

But EGFR was just a proof-of-principle target. The delivery system could, in theory, carry targeted siRNA to regulate any number of genes, including ones related to skin aging.

Mirkin?s enthusiasm for the possibilities of the new therapy is rare.?It?s unusual for a scientist to rave about his work in such terms, especially one in the field of siRNA, which has a checkered history filled with scientific frustrations and failed companies.

Ten years ago, scientists predicted a new era of siRNA therapies, but the hype machine ran into a roadblock: how to deliver siRNAs into cells and make them regulate the target gene and not cause any collateral damage.

The issue mainly has been size, Dowdy explained. Two million years of evolution have created cells that do a good job of keeping foreign stuff out. In order to enter a cell passively, a substance has to be about 1 nanometer or less, one reason why small molecule drugs -- pills -- are so tough to create. Another problem is that cell membranes carry a negative charge. They repel negatively charged molecules, like siRNAs.?

?It?s like trying to squeeze an elephant through the top of your desk,? Dowdy said.

Mirkin explained that his team?s delivery system relies on so-called ?scavenger receptors? on the cell surface to grab the nano SNAs and gobble them up. Once the SNAs were inside the cell, they were able to begin silencing the EGFR gene.

Dowdy said he?s ?cautious? about Mirkin?s claims for SNAs, given the history of other delivery systems that have been tried. So far no solution has been perfect, though siRNA technology is being tested in human clinical trials.?

Another issue is the nanospheres themselves. For example, the Food and Drug Administration has yet to rule on the use of nanoparticles in sunscreens until more data can be gathered on what effects, if any, the particles might have if they pass through skin and enter the bloodstream. Last week, the agency announced that the tiny technology needs more safety testing before it can be used in consumer goods.

But Dowdy did agree that for use on skin -- especially in cases like psoriasis or diabetic wounds where the skin surface has already been compromised, Mirkin?s group may be onto something.

Mirkin certainly hopes so. He?s founded a company called Aurasense Therapeutics. Just as Botox was originally meant to be used for muscle disorders, but now makes heaps of money thanks to its wrinkle-fighting properties, Aurasense stands to reap a windfall if anybody can walk up to a counter and buy a lotion or cream that really will reduce the signs of aging.

?I?ve never been more excited about anything,? Mirkin said.

Still, whether nano-delivered gene regulation is going to be the next Botox remains to be seen.
